Here, we isolated a novel bacterium (designated as strain TMPK1\u003cjats:sup\u003eT\u003c/jats:sup\u003e) from a 0.45-μm-filtered soil suspension by using a gel-filled microwell array device comprising 900 microwells and characterized its phylogenetic and physiological features. This strain showed low 16S rRNA gene sequence identities (\u0026lt;91%) and low average nucleotide identity values (\u0026lt;70%) to the closest validly described species, and belonged to a novel-family-level lineage within the order\u003cjats:italic\u003eRhodospirillales\u003c/jats:italic\u003eof\u003cjats:italic\u003eAlphaproteobacteria\u003c/jats:italic\u003e. Strain TMPK1\u003cjats:sup\u003eT\u003c/jats:sup\u003eexhibited small cell sizes (0.08–0.23 μm\u003cjats:sup\u003e3\u003c/jats:sup\u003e) and had a high cyclopropane fatty acid content (\u0026gt;13%), and these characteristics were differentiated from other\u003cjats:italic\u003eRhodospirillales\u003c/jats:italic\u003ebacteria. A comprehensive habitability search using amplicon datasets suggested that TMPK1\u003cjats:sup\u003eT\u003c/jats:sup\u003eand its close relatives are mainly distributed in soil and plant-associated environments. Based on these results, we propose that strain TMPK1\u003cjats:sup\u003eT\u003c/jats:sup\u003erepresents a novel genus and species named\u003cjats:italic\u003eRoseiterribacter gracilis\u003c/jats:italic\u003egen. nov., sp. nov. (JCM 34627\u003cjats:sup\u003eT\u003c/jats:sup\u003e= KCTC 82790\u003cjats:sup\u003eT\u003c/jats:sup\u003e).
