The novel strain is white when grown in liquid or solid media and forms Gram-negative rod-shaped, non-flagellated, non-motile cells. It conserves energy by aerobically oxidizing methane and hydrogen while deriving carbon from carbon dioxide fixation. Strain IT6\u003cjats:sup\u003eT\u003c/jats:sup\u003e had three complete \u003cjats:italic\u003epmoCAB\u003c/jats:italic\u003e operons encoding particulate methane monooxygenase and genes encoding group 1d and 3b [NiFe] hydrogenases. Simple carbon–carbon substrates such as ethanol, 2-propanol, acetone, acetol and propane-1,2-diol were used as alternative electron donors and carbon sources. Optimal growth occurred at 50–55°C and between pH 2.0–3.0. The major fatty acids were C\u003cjats:sub\u003e18 : 0\u003c/jats:sub\u003e, C\u003cjats:sub\u003e15 : 0\u003c/jats:sub\u003e anteiso, C\u003cjats:sub\u003e14 : 0\u003c/jats:sub\u003e iso, C\u003cjats:sub\u003e16 : 0\u003c/jats:sub\u003e and C\u003cjats:sub\u003e14 : 0\u003c/jats:sub\u003e, and the main polar lipids were phosphatidylethanolamine, aminophospholipid, phosphatidylglycerol, diphosphatidylglycerol, some unidentified phospholipids and glycolipids, and other unknown polar lipids. Strain IT6\u003cjats:sup\u003eT\u003c/jats:sup\u003e has a genome size of 2.19 Mbp and a G+C content of 40.70 mol%. Relative evolutionary divergence using 120 conserved single-copy marker genes (bac120) and phylogenetic analyses based on bac120 and 16S rRNA gene sequences showed that strain IT6\u003cjats:sup\u003eT\u003c/jats:sup\u003e is affiliated with members of the proposed order ‘\u003cjats:italic\u003eMethylacidiphilales\u003c/jats:italic\u003e’ of the class \u003cjats:italic\u003eVerrucomicrobiia\u003c/jats:italic\u003e in the phylum \u003cjats:italic\u003e\n                     \u003cjats:named-content content-type=\"phylum\"\u003e\n                        \u003cjats:ext-link xmlns:xlink=\"http://www.w3.org/1999/xlink\" ext-link-type=\"uri\" xlink:href=\"http://doi.org/10.1601/nm.31638\" xlink:type=\"simple\"\u003eVerrucomicrobiota\u003c/jats:ext-link\u003e\n                     \u003c/jats:named-content\u003e\n                  \u003c/jats:italic\u003e. It shared a 16S rRNA gene sequence identity of \u0026gt;96 % with cultivated isolates in the genus '\u003cjats:italic\u003e\n                     \u003cjats:named-content content-type=\"genus\"\u003e\n                        \u003cjats:ext-link xmlns:xlink=\"http://www.w3.org/1999/xlink\" ext-link-type=\"uri\" xlink:href=\"http://doi.org/10.1601/nm.32340\" xlink:type=\"simple\"\u003eMethylacidiphilum\u003c/jats:ext-link\u003e\n                     \u003c/jats:named-content\u003e\n                  \u003c/jats:italic\u003e' of the family '\u003cjats:italic\u003eMethylacidiphilaceae\u003c/jats:italic\u003e’, which are thermoacidophilic methane-oxidizing bacteria. ‘\u003cjats:italic\u003e\n                     \u003cjats:named-content content-type=\"genus\"\u003e\n                        \u003cjats:ext-link xmlns:xlink=\"http://www.w3.org/1999/xlink\" ext-link-type=\"uri\" xlink:href=\"http://doi.org/10.1601/nm.32340\" xlink:type=\"simple\"\u003eMethylacidiphilum\u003c/jats:ext-link\u003e\n                     \u003c/jats:named-content\u003e\n                  \u003c/jats:italic\u003e sp.’ Phi (100 %), ‘\u003cjats:italic\u003eMethylacidiphilum infernorum’\u003c/jats:italic\u003e V4 (99.02 %) and ‘\u003cjats:italic\u003e\n                     \u003cjats:named-content content-type=\"genus\"\u003e\n                        \u003cjats:ext-link xmlns:xlink=\"http://www.w3.org/1999/xlink\" ext-link-type=\"uri\" xlink:href=\"http://doi.org/10.1601/nm.32340\" xlink:type=\"simple\"\u003eMethylacidiphilum\u003c/jats:ext-link\u003e\n                     \u003c/jats:named-content\u003e\n                  \u003c/jats:italic\u003e sp.’ RTK17.1 (99.02 %) were its closest relatives. Its physiological and genomic properties were consistent with those of other isolated ‘\u003cjats:italic\u003e\n                     \u003cjats:named-content content-type=\"genus\"\u003e\n                        \u003cjats:ext-link xmlns:xlink=\"http://www.w3.org/1999/xlink\" ext-link-type=\"uri\" xlink:href=\"http://doi.org/10.1601/nm.32340\" xlink:type=\"simple\"\u003eMethylacidiphilum\u003c/jats:ext-link\u003e\n                     \u003c/jats:named-content\u003e\n                  \u003c/jats:italic\u003e’ species. Based on these results, we propose the name \u003cjats:italic\u003eMethylacidiphilum caldifontis\u003c/jats:italic\u003e gen. nov., sp. nov. to accommodate strain IT6\u003cjats:sup\u003eT\u003c/jats:sup\u003e (=KCTC 92103\u003cjats:sup\u003eT\u003c/jats:sup\u003e=JCM 39288\u003cjats:sup\u003eT\u003c/jats:sup\u003e).
